背景簡(jiǎn)介
自身免疫抗體是機(jī)體在非正常狀態(tài)下，由機(jī)體自身免疫系統(tǒng)分泌的針對(duì)機(jī)體自身蛋白的抗體。比如：系統(tǒng)性紅斑性狼瘡疾病產(chǎn)生的抗體。自身抗體產(chǎn)生的原因有很多，具體機(jī)理還有待研究。多數(shù)學(xué)者認(rèn)為：一些自身抗體的產(chǎn)生是由于遺傳因素和環(huán)境因素共同作用的結(jié)果，比如：一些病毒性疾病或長(zhǎng)期在處于有毒化學(xué)毒素下，都可以導(dǎo)致自身免疫疾病。自身免疫疾病源與機(jī)體對(duì)自身體內(nèi)或組織產(chǎn)生免疫應(yīng)答。換句話說，免疫系統(tǒng)將機(jī)體自身的物質(zhì)誤認(rèn)為病原。這主要局限于一些特定組織：（比如：甲狀腺）。自身免疫疾病的治療主要依賴于免疫調(diào)節(jié)藥物用于降低免疫應(yīng)答。
自身免疫疾病對(duì)機(jī)體的傷害程度取決于自身抗體作用的的器官或組織和器官特異性的自身抗體導(dǎo)致的紊亂程度。那些只對(duì)單一器官作用自身抗體導(dǎo)致的自身免疫疾病很容易發(fā)現(xiàn)，因?yàn)檫@類疾病常常伴有系統(tǒng)的病癥。比如：甲狀腺機(jī)能亢進(jìn)和橋本氏甲狀腺炎。由自身免疫抗體導(dǎo)致的系統(tǒng)性功能紊亂，往往很不易被察覺。自身抗體可通過ELISA 方法檢測(cè)?；颊呖赡苡嗅槍?duì)不同抗原的自身抗體。用于自身免疫疾病研究的動(dòng)物模型有很多。已經(jīng)發(fā)現(xiàn)的：18個(gè)自身免疫基因簇中有9個(gè)基因簇與相應(yīng)的動(dòng)物模型在序列上高度保守。動(dòng)物模型被廣泛應(yīng)用于人自身免疫疾病的研究，包括：多發(fā)性硬化癥、糖尿病、風(fēng)濕性關(guān)節(jié)炎、系統(tǒng)性紅斑狼瘡。通過人為誘導(dǎo)、自然的以及遺傳操作的各種動(dòng)物模型被用于研究自身免疫疾病和相應(yīng)的藥物研究。
