Hypoxyprobe公司基于缺氧探針抗體技術(shù)，在此技術(shù)上開發(fā)了系列缺氧探針標(biāo)記試劑盒，為組織疾病的診斷和治療提供有力研究工具。
檢測(cè)原理
低氧標(biāo)志物測(cè)定是利用還原硝基有選擇性結(jié)合乏氧細(xì)胞的能力而形成的一種從細(xì)胞水平測(cè)定腫瘤乏氧的技術(shù)，其原理是：腫瘤乏氧細(xì)胞還原能力強(qiáng)，當(dāng)具有電子親合力的硝基咪唑主動(dòng)擴(kuò)散透過細(xì)胞脂膜，在細(xì)胞內(nèi)硝基還原酶作用下，硝基被還原，還原產(chǎn)物與大分子物質(zhì)不可逆結(jié)合，滯留在組織內(nèi)。而正常含氧細(xì)胞親電子硝基咪唑還原產(chǎn)物立即被氧化，從而用硝基咪唑類化合物在腫瘤內(nèi)的代謝程度來反映腫瘤的乏氧
Pimonidazole是新型的乏氧細(xì)胞標(biāo)記物，HPI公司擁有硝基咪唑類化合物特異性抗體，該抗體為公司技術(shù)，可用于免疫組織化學(xué)、酶聯(lián)免疫吸附試驗(yàn)和流式細(xì)胞分析檢測(cè)。
Pimonidazole 應(yīng)用
特異性的低氧探（Hypoxyprobe-1）zui初用于反映腫瘤細(xì)胞內(nèi)的低氧梯度，正常細(xì)胞無表達(dá)或低表達(dá)。而在低氧的腫瘤細(xì)胞內(nèi)可見高表達(dá)，并且隨低氧的程度越重而顏色越深。由于其很好的化學(xué)穩(wěn)定性、水溶性，同時(shí)小分子的探針可以自由穿過血腦屏障而到達(dá)腦組織細(xì)胞及腦腫瘤細(xì)胞。在實(shí)驗(yàn)中應(yīng)用低氧探針可以形象直觀的通過免疫組化來分析腦挫傷周圍組織細(xì)胞低氧的范圍和程度，同時(shí)對(duì)實(shí)驗(yàn)設(shè)備及器材要求較低，為無創(chuàng)性實(shí)驗(yàn)。
